Recently, Xgene Pharmaceutical Inc. (“Xgene”), a company focusing on the research and development of innovative drugs in the fields of pain and neurology, completed a RMB 250 million Series D financing round. This round was jointly led by Yifeng Capital and Gaotejia Investment, with participation from Jiaxing Yuanfan, Real Capital Global, and other institutions. Kaicheng Capital served as the exclusive financial advisor for this financing. The funds raised will primarily support the Phase III acute pain clinical trial of its core product, XG005, in the United States, while also advancing the development of its entire pipeline and its global commercialization strategy.
The global analgesia field has long faced clinical challenges: while traditional opioids offer clear analgesic effects, the risks of addiction, abuse, and adverse reactions have long plagued clinical practice. Globally, there is a significant unmet need for innovative non-opioid drugs with potent analgesics and improved safety. Changyu Pharmaceuticals is focusing on this area, leveraging its proprietary dual-effect drug conjugation technology platform to build a complete pipeline covering acute, chronic, and localized pain, and developing oral and transdermal delivery methods, aiming to develop innovative solutions that can reduce or even replace opioids.
The core candidate drug, XG005, is a globally innovative oral non-opioid dual-mechanism analgesic small molecule that simultaneously inhibits inflammatory and neuropathic pain signaling pathways. Previous Phase II/III studies in the United States yielded positive clinical results, demonstrating good efficacy and safety in the indication of postoperative acute pain. Following this round of financing, based on the completed Phase 2b/3 studies, the drug will advance to Phase III clinical trials, aiming for a key milestone in overseas market launch. Simultaneously, multiple clinical trials for different pain indications are being conducted domestically.
Capital continues to pour into the pain management sector, driven by a clear unmet medical need. Pain is a broad indication spanning surgery, orthopedics, and oncology, offering vast market potential. However, innovative drug development is challenging, and the number of domestic Chinese companies developing differentiated non-opioid analgesics is relatively small. Investors Yifeng Capital and Gaotejia both stated that they are optimistic about the long-term clinical demand in the pain management field, recognize Xgene’s outstanding R&D team and differentiated technology platform, and are optimistic about the company’s future development potential in the global analgesia market.
From an industry perspective, the domestic analgesic market in China is currently dominated by generic drugs and traditional analgesics, with a scarcity of new-generation non-opioid innovative drugs. As public awareness of pain management increases and countries strengthen opioid control, innovative analgesics with novel mechanisms and low addiction risks have a promising long-term market prospect. Meanwhile, overseas markets have strong demand for novel analgesics, and innovative pharmaceutical companies with global simultaneous development capabilities are more likely to receive valuation premiums.
For Xgene, the completion of its Series D financing marks a crucial stage in validating clinical value. Phase III clinical trials for new drugs require significant investment, and the clinical data results will directly determine the product’s commercialization prospects. In addition to its core product XG005, the company continues to enrich its pain management pipeline, building a product portfolio for long-term sustainable development.
Pain and central nervous system (CNS) innovative drugs represent a high-barrier-to-entry research and development field, with a lack of candidate drugs successfully advancing to Phase III clinical trials. This funding largely facilitates Xgene to accelerate its global clinical development process. Experts in the field will continue to monitor the overseas Phase III clinical trial readout for XG005, as well as the company’s progress in domestic and global commercialization.
